Translating Polygenic Analysis for Prevention
نویسنده
چکیده
There are no easy answers in complex disease genetics. Common, chronic health conditions such as obesity and heart disease are influenced by many genetic variants scattered across the genome, with each variant making small contributions to risk. Ever-larger genome-wide association studies (GWAS) are uncovering more and more of these variants. For many diseases and disease-related traits, discovered loci now number in triple digits. As GWAS sample sizes grow, the volume of discovered variants is expected to increase further. Translation of this growing volume of genetic discoveries to improve population health is needed.

Genes Predisposing to Monogenic, Polygenic, and Syndromic Obesity: A Review of Current Trends and Prospects for Standard Obesity Genetic Testing
Objective: The burden of obesity is currently enormous, necessitating a novel strategy to complement the existing ones. Accordingly, genetic predisposition is suspected in many cases of the disease, which can potentially be used as therapeutic targets. However, there are differing viewpoints on the suspect genes, prompting the current review to articulate the genes and their mechanisms. Eight (...

At the present study, the observations of performance traits including body weight at 8 and 12 weeks of age, age at sexual maturity, egg weight and egg number were analyzed to detect major genes in Yazd native fowl. The probability of segregation for major genes was studied using simple tests including tests of normality, Bartlett, Levene and Fain for residuals and Bayesian analysis. Segregati...
متن کاملType 2 diabetes heritability in the Tehran families: Tehran cardiometabolic genetic study
Introduction: Type 2 diabetes, as a complex disorder, is one of the most prevalent endocrine disorders. Importantly, the extent of familial aggregation and heritability in Iran is unknown. The aim of the present study was to determine type 2 diabetes heritability in the Tehran families. Materials and Methods: The current research comprises 1691 diabetic and 12050 non-diabetic individuals over ...
متن کاملPublic health genomics and personalized prevention: lessons from the COGS project
Using the principles of public health genomics, we examined the opportunities and challenges of implementing personalized prevention programmes for cancer at the population level. Our model-based estimates indicate that polygenic risk stratification can potentially improve the effectiveness and cost-effectiveness of screening programmes. However, compared with 'one-size-fits-all' screening prog...
متن کاملPolygenic Risk Score Identifies Subgroup With Higher Burden of Atherosclerosis and Greater Relative Benefit From Statin Therapy in the Primary Prevention Setting.
BACKGROUND Relative risk reduction with statin therapy has been consistent across nearly all subgroups studied to date. However, in analyses of 2 randomized controlled primary prevention trials (ASCOT [Anglo-Scandinavian Cardiac Outcomes Trial-Lipid-Lowering Arm] and JUPITER [Justification for the Use of Statins in Prevention: An Intervention Trial Evaluating Rosuvastatin]), statin therapy led ...
